Antibacterial activity effect of Bacillus velezensis AYGS-17 extract against periodontal and cariogenic pathogens

Abstract

|
Strain AYGS-17, a microorganism with antibacterial activity against periodontal pathogens, such as Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans and cariogenic pathogens, such as Streptococcus mutans, was isolated in culture. Through genetic analysis using average nucleotide identity, the isolate was identified as Bacillus velezensis, with a similarity of 98.72% to Bacillus velezensis NRRL B-41580. The optimal growth of B. velezensis AYGS-17 was observed on yeast extract?peptone?dextrose (YPD) medium at 25¡ÆC and pH 6.5, and the highest antibacterial activity was observed in tryptic soy broth (TSB) medium. Therefore, strain AYGS-17 was cultured on TSB medium at 25¡ÆC and pH 6.5 for 48 h. Subsequently, 100% methanol was used to extract a natural compound which was tested against periodontal and cariogenic pathogens to determine its minimum inhibitory and bactericidal concentrations. Porphyromonas gingivalis, A. actinomycetemcomitans, and S. mutans were inhibited and killed at concentrations of 10, 25, and 50?100 ¥ìg/ml, respectively. In conclusion, the natural extract from strain AYGS-17 possesses potential commercial value for applications in the development of food, drugs, and other products because it effectively inhibits periodontal and cariogenic pathogens.
